The swallowing system is comprised of a single tube

Each system contains a series of valves which change
the shape or configuration of the system or protect it.
Valves created by the lips and tongue keep food in the
mouth and in place prior to swallowing. The valve
created by the cricopharyngeous muscle at the top of
the esophagus keeps air out of the digestive system
during breathing. The valve created by the soft palate
keeps food out of the nasal airway during swallowing.
The valves created by the false and true vocal folds and
the epiglottis prevent food from entering the airway
during swallowing.
Shared Pathways Exist between the Feeding and
Respiratory Systems
The feeding and respiratory systems share a portion of
the pharynx between the area behind the tongue and
the area at the entrance to the larynx and the entrance
to the esophagus.

Source text - English Introduction
During embryonic development Shh acts as a morphogen
in several different tissues [1,2]. The vertebrate neural
tube is a particularly well-studied example and significant
progress has been made in identifying Shh target genes
and analyzing the mechanism by which cells interpret
Shh signalling [3,4]. At early stages of its development,
the neural tube consists of proliferating neural progeni-
tors. Shh secreted from the notochord and floor plate, at
the ventral midline of the neural tube, forms a ventral to
dorsal gradient that is interpreted by progenitors to estab-
lish patterns of expression of homeodomain (HD) and
basic helix-loop-helix (bHLH) transcription factors
(TFs). The combinatorial expression of these genes
divides the ventral neural tube into discrete dorsal–ventral
domains of progenitors, each of which generates
specific subtypes of motor neurons and interneurons
(Figure 1a,b).
Shh is transduced through a signalling pathway that
ultimately controls the activity of the Gli family of
sequence specific DNA binding TFs [1,5]. In the absence
of Shh, Gli proteins are either degraded or converted into
transcriptional repressors (GliR) by partial proteolytic
processing. Shh signalling blocks this processing and
generates activator Gli isoforms (GliA). Thus, in the
neural tube the Shh ligand gradient is transduced into
opposing gradients of GliA and GliR (Figure 1c). Evidence
from mice lacking one or more of the three Gli
genes that make up the family indicates that Gli2 predominantly
contributes transcriptional activator function
and only weakly acts as a repressor [6,7]. By contrast, Gli3
acts principally as a repressor and only has a minor
activator role [8]. Gli1 is induced by Shh signalling and
acts solely as a transcriptional activator; however, neural
tube development is unaffected by the deletion of the
Gli1 gene, suggesting that it plays little if any role in this
tissue [9]. Beyond these initial observations, a key question
is how the Shh–Gli gradient regulates differential
gene expression in the neural tube and several recent
studies have begun to provide insight into the underlying
molecular logic [10,11,12].
Binding site affinity and the regulation of Shh
target genes
In conventional models of morphogen patterning, which
we will refer to as Affinity-Threshold models [13], there is
a positive correlation between the range of gene induction
and the affinity of binding sites within cis-regulatory
modules (CRMs) of morphogen regulated TFs. CRMs of
genes activated by low morphogen thresholds (at long
range) have a higher binding affinity for morphogen
regulated TFs than those activated by high morphogen
concentrations (at short range). However, this model does
not readily accommodate bimodal TFs such as Gli, which
act as both repressors and activators, since changing
binding site affinity will affect binding of both isoforms
[14]. Indeed analysis of Gli Binding Sites (GBSs) located
within CRMs of Shh target genes in the neural tube did
not find a positive correlation between the binding affinity
and the range of gene induction [11]. Instead, some
CRMs containing high affinity GBSs are associated with
short-range induction (e.g. Nkx2.2), whereas others
associated with genes expressed at a greater range contain
GBSs with low binding affinity (e.g. Nkx6.1) [11,12].
